No, you can not switch to automatic from manual. How can you get tired from driving stick? I drive manual everyday and I have no problem with getting tired.
The Automatic in the S5 is tiptronic, meaning you can switch from automatic to manual, but the manual is different from the standard manual and the Tiptronic Automatic is slower than the Manual.
The S5 is a 2 door sports coupe and the E class is a luxury sedan that competes with the Audi A6. The S5 is much faster, much better handling, much safer, and they have the same luxury. The S5 is also more powerful. It is a closer match to the AMG Mercedes E class, which is the sports version of the E-class.

Dude, the Audi S5 is awesome. And where in the world did you get the idea that you could just switch a car from manual to automatic? The closest thing is when you get the automatic it comes with tiptronic so that you can manually switch the gears, but it’s just not the same as having a manual transmission since you don’t have a clutch, it’s the lazy manual.
If you’re buying it, seriously, go with the manual, I drive a manual car, and i hate it every time i drive my mom’s automatic, it’s just not the same. Manual is so much better.
And as far as performance, I don’t know the exact figures between the two. But, I’d take the Audi over the Mercedez any day. I think Mercedez are overrated, and the S5 has a lot of power, and it’s a really nice car. I would love to own one. But basically, I’d take an Audi over any Mercedez (except the SLR McLaren) (but, I’d take an R8 or RS4 over the SLR) haha. But yeah. The S5 is sweet.
